Welcome to "Fluorescence Microscopy of Wood". This multimedia CD contains methods for conventional and confocal fluorescence imaging of wood cell walls and will be of interest to wood anatomists, wood technologists, and plant biomaterials researchers. We describe methods for acquiring autofluorescence images and images from stained sections of wood using Arabidopsis, bamboo, Eucalyptus, fern, poplar, radiata pine, tobacco, and wheat xylem as examples. We also provide chemical and spectral information for the following dyes; Acriflavin, basic fuchsin, calcofluor, Congo red, safranine, Schiff’s reagent and toluidine blue. Also included are examples of reflectance imaging and 3D projections including anaglyphs and 3D movies, as well as tutorials on confocal microscopy and digital image processing. The CD includes over 500 Mb of 3D movies and more than 120 high resolution images of wood structure.
